how can i get end effector velocity from joint velocities ?
What does the Rotation matrix mean ?

You should learn about the kinematics of robot manipulator for basic. and then we can use this basic to calculate the end effector velocity by using Jacobian matrix.

The Jacobian matrix is somewhat complex math that I always had trouble with when I was a student . . . and you first need to calculate your inverse kinematics of your robot arm to use it, a challenge itself! It's possible, for certain limited motions, to calculate end effector velocity with simple algebra.
If you google around, you should be able to find equations to help you out.
